The effect of antisense c-myc by liposomes on the growth of human hepatoma in nude mice

摘要

Objective: To investigate the effect of LR mediated c-myc antisense oligodeoxynucleotides (ASODN) on the growth of human hepatoma cell SMMC-7721 in nude mice, in which were identified overexpression of c-myc gene. Methods: After 1.5 × 106 human hepatoma cells SMMC-7721 were inoculated into each backside skin in 9 nude mice, all nude mice developed tumor node on day 14. Then we divided them into 3 groups: experiment, empty control and mismatch control. We injected directly 10 μmol·L-1 LR entrapped ASODN into the tumor body of the experiment group, MODN or water into control group for 5 days, and observed for 28 days Results: By measuring tumor body size, we found that the size of group treated with ASODN was significantly different from that of control at day 5 (P<0.05). The weight of tumor between the two groups had significant difference (P<0.05) on day 28. There were differences in histology by HE stains. Immunohistochemical detection showed that C-myc protein expression was decreased in experiment group. Conclusion: LR entrapped ASODN may inhibit the growth of human hepatoma in nude mice by direct injection.
