Aim: To investigate the protective effect of major constituents in total panax notoginsenoside (TPNS) on rat brain microvessel endothelial cell (RB-MEC) damage induced by H2O2. Methods: RBMEC was successfully isolated and primarily cultured. The protective effect of TPNS, R1, Rg1, Rd, Re and Rb1 on damaged RBMECs was investigated by means of MTT and lactic dehydrogenase (LDH) in the layers of RBMEC. Results: H2O2 damaged RBMEC markedly at the concentration of 500 μmol·L -1. The results of MTT and LDH showed markedly protective effect on damaged RBMEC at the concentration of 20-200 mg ·L-1 for TPNS, 16.0 μmol·L-1 for R1, 37.5-75.0 μmol·L -1 for Rg1, 8.0-16.0 μmol·L-1 for Rd and 5.4-54.0 μmol·L-1 for Rb1 respectively(P < 0.05 or P < 0.01). Re showed no markedly protective effect. Conclusion: TPNS showed markedly protective effect on primarily cultured RBMEC damage induced by H 2O2. Ginsenoside Rb1, Rg1 and Rd are probably the major constituents in TPNS which play important role in the brain protection.
