Influence of several experimental factors on morphine dependence model in rats

Objective: To study the influence of several experimental factors on morphine dependence in rats. Methods: The 130 SD health rats were randomly assigned to control groups and dependence groups. The withdrawal syndromes between control groups and dependence groups in different dosage, different time, constant dose administration and increased dose administration were evaluated. Results: The differences between control groups and dependence groups were significant (P<0.05) in DG₂ and DG₃, and the differences between DG₂, DG₃ and DG₁ were also significant (P<0.05) in the same dependence groups. In 3 d, 5 d and 7 d groups, the differences between control groups and dependence groups were significant (P<0.05) in 7 d, and the differences between 3 d, 5 d and 7 d were significant (P<0.05) in the same dependence groups. The differences between control groups and dependence groups were all significant (P<0.05) in constant groups and increased groups, and the differences between constant groups and increased groups were also significant (P<0.05) within the same dependence groups. Conclusion: Morphine dependence forming process is related to several experimental factors, including different dosage, different time, constant dose, and increased dose. Therefore, more attention should be paid to these experimental factors in setting up morphine dependence model and choosing the best experimental methods.
