摘要

Objective: To explore the correlationship between topiramate treatment and the expression and activity of acetylcholine esterase (AChE) in mouse footpad sweat gland. Methods: Topiramate was given to two-weeks-old mice at 80 mg/kg for 1 month. By immunohistochemical staining and Western blot, the expression of AChE was analyzed in mouse footpad sweat gland in topiramte treatment group and non-treatment group. The AChE activity was determined by the method of Ellman with some modifications. Results: By the immunostaining, AChE optical density value in the topiramate treatment group and control group was 1.09±0.03 and 0.98±0.09, respectively. There was no statistical difference (t=0.572, P>0.05). No significant difference of AChE expression was found between these two groups by Western blot (t=0.394, P>0.05). The AChE activity was (1.42±0.38) μmol/mg and (1.37±0.42) μmol/mg, respectively in treatment and control groups (t=0.746, P>0.05). Conclusions: The expression and activity of AChE in mouse footpad sweat gland are not affected by topiramate treatment.
