摘要
Objective: To explore the effects of small interfering RNA (siRNA) knockdown of forkhead box M1 (FoxM1) on the invasiveness of human lung cancer cell A549 in vitro so as to provide theoretical evidence for gene-targeted therapy for lung cancer. Methods: FoxM1 siRNA was transfected into human lung cancer A549 cells with lipofectamine 2000. Protein expression level was evaluated by Western blotting. Anchorage-independent growth and invasiveness of A549 cells were measured by soft agar colony formation and transwell cell invasion assay, respectively. Results: FoxM1 siRNA was successfully transfected into A549 cells, resulting in the significant inhibition of FoxM1 mRNA and protein expression. Down-regulation of FoxM1 decreased the invasiveness of A549 cells and reduced the expression of matrix metalloproteinase-2 (MMP-2) of lung cancer cells. Conclusion: Down-regulation of FoxM1 can inhibit the invasiveness of human lung cancer cells, suggesting that FoxM1 is a potential target for lung cancer therapy.
