Abstract
Objective: To study the fabrication methods of scaffold with 3D bone microchannel structure and culture in vitro after seeding osteoblast. Observing osteoblastic growth, proliferation and differentiation in the 3D porous scaffolds. Method: The rapid prototyping (RP) technology was used to fabricate negative molds of scaffolds. CPC pasty was cast into the molds. Subsequently, the molds were fired out after CPC being hardened to obtain the scaffolds with 3D bone microchannels. Osteoblast cells were seeded and cultured in vitro. Samples were taken out at the 4th day and 14 day and observed using scanning electron microscope. Results: The scaffolds indirectly fabricated by Stereolithography RP technology obtained good 3D spatial structure. Osteoblast can attach and proliferate on the surface and in the micropore of scaffolds. Conclusion: It is feasible for indirectly fabricating 3D porous scaffolds using RP technology in the application of bone tissue engineering. The CPC scaffolds construct provided an acceptable scaffold for osteoblastic growth, proliferation and differentiation.
