Abstract
Cluster of differentiation 147 (CD147) is pivotal in the onset and progression of tumors. Nevertheless, there is a dearth of small-molecule drugs that specifically target CD147 in the clinic. To address this, a stable bionanomaterial, which integrates cell membranes containing CD147 with magnetic zeolitic imidazolate framework-90 (Fe3O4@ZIF-90), was ingeniously designed and synthesized for active ingredient screening in this work. The Fe3O4@ZIF-90@CD147 bionanomaterial was obtained by specifically immobilizing SNAP-tagged CD147 cell membrane fragments (CD147-ST) onto the surface of BG (benzylguanine)-modified Fe3O4@ZIF-90 particles. Characterization findings indicate that this bionanomaterial exhibits outstanding adsorption capacity along with exceptional specificity and selectivity for target molecules. The bionanomaterial was then applied in CD147-targeted antitumor drug leads screening from Ophiopogon japonicus (L.f.) Ker-Gawl extracts. Two hit compounds, methylophiopogonanones A (MOA) and B (MOB), were obtained. Additionally, preliminary pharmacological experiments demonstrated that both the MOA and the MOB exhibited inhibitory effects on the proliferation and migration of Huh-7 cells.
