Abstract
Objective: To study the effects of miRNA-449a/b on apoptosis of gastric cancer cells and expression of its target CCNE2. Methods: The expression of miRNA-449a/b in gastric cancer cell line BGC823 after transient transfection was detected by Real-time PCR. Flow cytometry was used to detect cell apoptosis rate. The target gene of miR-449a/b was detected by Western blot. Results: At 48 and 72 h, miRNA-449a/b was overexpressed in transient transfection cells and the upregulated miRNA-449a/b could inhibit proliferation and promote apoptosis (P<0.05). The expression of CCNE2 was also inhibited significantly (P<0.05). Conclusion: miRNA-449a/b inhibits gastric cancer cell probably by targeting CCNE2.
