Abstract
This study was aimed to expolore the correlation between effects of lactacystin and arsenic trioxide on the rpt-1 subunit (S7 subunit) and β1 subunit of 26S Proteasome in NB4 cells. NB4 cells were treated with lactacystin, AS2O3, or combine-treated with lactacystin plus AS 2O3 for 24h and 48h. Performed western blot assay to evaluate the effect of AS2O3 and lactacystin on expression of β1-subunit protein and rpt-1 subunit(S7 subunit) protein. Combine-treated could induce β1-subunit protein expression increase after 24h or 48h. Moreover, agents could induce rpt-1 subunit protein expression decrease after 24h, but couldn't induce it after 48h. It could be indicated that to induce β1-subunit protein increase lactacystin and AS2O 3 have a synergistic effect. However, don't have a synergistic effect to induce rpt-1 subunit decrease.
