Effect of survivin on choriocarcinoma cell JAR apoptosis induced by 5-fluorouracil

Abstract

Objective: To study the role of survivin in choriocarcinoma cell JAR apoptosis induced by 5-fluorouracil (5-Fu). Methods: The choriocarcinoma cell JAR strain was chosen in this study. The inhibitory effects of chemotherapeutical drug 5-Fu on JAR strain were assayed with MTT method. Apoptosis was detected by AnnexinV/PI test. Expression of survivin was detected by reverse transcriptase PCR and Western blot. Results: Circled digit one 5-Fu inhibited the JAR cell growth in a dose-and time-dependent manner. Circled digit two 5-Fu caused the apoptosis of JAR cell in a concentration-dependent manner. Circled digit three Survivin gene and protein decreased with the increase of 5-Fu. Conclusion: The decrease of the expression of survivin may play a critical role in JAR cell apoptosis induced by 5-Fu.
